By binding to the NMDA receptor with a higher affinity than Mg2+ ions, memantine is able to inhibit the prolonged influx of Ca2+ ions, particularly from extrasynaptic receptors, which forms the basis of neuronal excitotoxicity.

“Memantine was started at 5 or 10 mg QHS, before being increased by 5 or 10mg every 4-7 days, as tolerated, to a maximum dose of 40 mg - 60 mg, as tolerated. In all, 13 patients showed complete remission from CRPS with VAS 0 and the disappearance of allodynea for at least nine months after the use of memantine.

Memantine isn't very long term memory impairing, in comparison to DXM, or even Ketamine, but short term memory does seem to suffer a bit and you can lose trains of thought quite easily at high doses. It's clean feeling, compared to DXM.

Memantine is great. It is a long acting NMDA receptor antagonist that binds to an open channel binding site. This means that for memantine to bind, the neuron has to depolarize, and open to clear the magnesium block. Then memantine is pulled into the channel to its binding site.

2015年6月15日 · Memantine is obviously one of the safer NMDA antagonists, but it has still been noted to cause vacuolation and neuronal damage at high doses in rodents. The toxic dose in humans is not known, but extrapolating from the therapeutic-to-toxic ratio in rodents, you are definitely flirting with this toxicity.

2016年3月25日 · Honestly, I think it's because Memantine + nicotine is a bad combo, especially for those (like me) who are addicted to the habituating effects of cigarettes. Memantine is a nAchr antagonist, which feels..unpleasant, if you smoke regularly. This is only my theory.
